Avoid defeat and they will be in the second round draw, which takes place on ITV4 on Monday after Dagenham & Redbridge v Salford City.
The U’s have only fallen at the first round twice in the last nine seasons, while Banbury have never got past this stage.
Victory this weekend is worth £22,629 to each club, but the Puritans have already banked considerably more from this season’s Cup run.
They have gained £20,625 in prize money just by reaching the first round and net a broadcast fee of £50,000 from Saturday’s game.
