Ross Butler, Rob Powell and an own goal made up Drayton's tally, with Luke Regan, Liam Humphries and Marcus Richards on target for Coleshill.
They are level on points with Harwell Village, but have a superior goal difference.
Crowmarsh Gifford put a serious dent in Coleshill Res' Division 2 title ambitions with a 3-1 success.
Phil Hedges led the way with a hat-trick, with Danny O'Driscoll also netting on his return following a six-month injury lay-off.
Goals from Johnny James and Chris Houlier earned Grove Rangers a 2-1 home win against Saxton Rovers Res.
In Division 3, Didcot Casuals moved into the top three with a 3-0 win at Benson Lions with strikes from Jamie Newport, Matty Evans and an own goal.
Stanford-in-the-Vale edged to a 3-2 home win against Marcham Res.
Ben Harrison, Daniel Bradbury and Jamie Hopgood were the Stanford marksmen, with Leighton Jackson and Declan Fulton scoring for Marcham.
In Division 4, Eddie Yarker and Joe Schofield were on target for Harwell Village Res as they drew 2-2 with Uffington United.
Garry Haines and Gareth Armishaw netted for Uffington.
Benson AFC Res won 4-1 at Hagbourne United Res in Division 5 to make up ground on the leaders Coleshill A, who were without a game.
Carl Fisher, Teddy Carter, Jamie Martin and Tim Millard found the net for Benson.
Faringdon Town A thrashed Uffington United Res 5-1, thanks to a hat-trick from Ben Henley his first of the season.
James Tyler and Andy Dunbar got the others, with Darren Morton replying.
Jamie Clark hit a double for Harwell International A as they beat Kennington Res 4-1.
Rob Johnson and Steve Cummings completed their tally.
Steve Pitson scored the late winner for Saxton Rovers as they saw off Childrey 1-0 in the first round of the North Berks Cup.
AG Kingham Cup holders Shrivenham A are out of the competition at the second-round stage after a 1-0 defeat against Lambourn Sports Res. Terry Amor got the winner.

Ardington & Lockinge had to endure extra time before beating ten-man Wootton & Dry Sandford Res 6-3.
Martin Clarke, Joffy Boreham and Jon Fisher put Wootton into a 3-0 lead, before they were reduced to ten men.
A Matt Searis hat-trick and strikes from Frankie Dorrian, Paris Lewis and Sam Powell secured victory for Ardington.
