Manager Kevin Brock says Banbury United need to toughen up as they travel to Division 1 South & West outfit Marlow in the second qualifying round tomorrow.
Brock was frustrated last week as Banbury threw away a 2-0 lead to lose 3-2 at home to Rugby United, and he said that individual defensive errors are costing them dear.
Brock said: "We've been looking OK, but at this level, individual mistakes cost us.
"Sometimes you get away with them, but we're having a run just lately where we're being punished for most of them.
"We've lost at home to Gloucester and Rugby recently, and so it's been very frustrating."
Brock is hoping that tomorrow's tie will bring a positive response from his players, with the Conference clubs due to enter the competition the round after next.
"The Trophy is the premier competition for non-league football, and we would certainly benefit from a good run in it," he added.
Jon Gardner and Andy Baird are carrying hamstring injuries, but are both expected to come through fitness tests.
Meanwhile, defender Daniel Szczukiewicz is back in Poland this weekend.
Brock added: "I'm looking for a positive response from the players."
Didcot Town face a tricky trip to Southern League Premier Division outfit Chippenham Town.
Manager Stuart Pearce has a number of injury concerns, with Ian Concannon (hamstring) the most doubtful.
Stuart Beavon (ankle) is also struggling, while Dave Green (hamstring) and Jamie Brooks (groin) face fitness tests.
On-loan Andrew Gunn is unavailable as Oxford United will not allow Didcot to play him in the Trophy, while Grant Goodall misses the trip due to family commitments.
Manager Stuart Peace said: "This will be a good distraction from the league and will be an excellent test of how far we've progressed and what we need to do to step up to another level."
Abingdon United also face a trip to a Southern League Premier Division side in the shape of Hemel Hempstead Town.
Richard Peirson returns from injury, as does Ryan Curtin from illness. However, Ryan Brooks is suspended.
