Letcombe's unbeaten run of 18 games came to an end in the last four minutes at Prestwood in the second round of the Challenge Cup.
The visitors wasted a string of chances in a goalless first half, while keeper Stuart Game had to save well from Matt Cox to keep his side on terms.
On the hour, a low shot from Robbie Fairchild crept under the dive of keeper Adam Slattery to put Letcombe ahead, but Prestwood equalised five minutes later through Richard Pontin.
Letcombe seemed to have won the game four minutes from time when substitute Liam McCullough netted from a narrow angle.
However, Prestwood equalised with a superb goal straight from the restart through Scott Hawkes and the winner came in injury time.
Letcombe failed to clear a corner, and Pontin headed in from two yards.
North Leigh 0,
Wantage Tn 3
Division 1 East leaders Wantage fully deserved their Supplementary Cup first-round win at Premier Division champions North Leigh.
The home side appeared more preoccupied with next week's FA Vase tie than with the job at hand, although this should take nothing away from Wantage, who have come on leaps and bounds under manager Andy Lyne.
After a goalless first half in which North Leigh's Matty Shelton had his penalty saved by Wantage keeper Paul Belcher, the visitors went in front in the 57th minute with a breakaway goal from Adie Tinsley.
Six minutes later, Wantage went further ahead from a JP Mills penalty after his initial shot was handled on the line.
North Leigh introduced three youngsters into the fray, but Wantage made it 3-0 in the 75th minute when Gary Swann wrapped up the victory from Glyn Jones's free-kick.
Abingdon Utd 1,
Slimbridge 2 (aet)
United fielded new signings Ani Odhiambo and Julian McCalmon from Quarry Nomads and even though the result did not go their way, it was another useful performance.
Abingdon forced Slimbridge onto the defensive in the first quarter, Odhiambo having a shot turned round the post on 25 minutes. From the corner, Mark O'Hara's drive was blocked by a defender.
After 32 minutes, McCalmon mis-hit a pass straight to Freeman, who was about to shoot when McCalmon got back and brought him down. Cole put Slimbridge ahead from the spot.
After the break, O'Hara's penalty was blocked by Chandler, but he followed up to level the score.
Odhiambo had a cracking effort tipped around the post by the keeper, before Slimbridge grabbed the winner in the 105th minute through Mike Green.
Milton Utd 3,
Witney Utd 1
Milton produced an excellent performance see off high-flying Witney.
Wide men Simon Kenny and Rob Powell kept the visitors' defence busy throughout and it was Powell who produced the cross for Simon Kenny to put the home side ahead on 11 minutes.
Witney came back when Ben Border swung over a free-kick on 31 minutes for Darren Teggart to head home the equaliser at the far post.
Milton regained their lead after the break. Paul Davis found Graham Edney, who cut inside and fired home from 18 yards.
A superb save by keeper Craig Griffiths kept Milton ahead, and Paul Davis sealed victory with fierce 20-yard drive.
Ardley Utd 3,
Quarry Nomads 1
A weakened Quarry side never got into this niggly cup tie.
It took only four minutes for Paul Bough to set up his brother Charlie, who put Ardley ahead.
Nigel Swafford made it 2-0 on the half-hour when he netted from an acute angle, but Quarry got back into the game when Ardley keeper Martin Woodward brought down John McMahon, who picked himself up to score from the spot.
The home side replied immediately with a four-man move culminating in a tap-in from Faruk Yarabi, before the game deteriorated into a succession of fouls which led to Quarry skipper Liam O'Callaghan receiving the red card.
