The leader of South Oxfordshire District Council has vowed to oppose any move to redraw part of Oxford's boundary.
Oxford City Council has asked the Boundary Commission to review the boundary south of Grenoble Road in Greater Leys.
Transferring Green Belt land in South Oxfordshire to the city council would make it easier to build thousands of new homes on the site.
But SODC leader Ann Ducker said: "We are totally opposed to development on this Green Belt land in South Oxfordshire.
"Oxford City Council wants to expand the city into south Oxfordshire without justification.
"The city should use underdeveloped land within its own boundaries to build housing, instead of trying to commandeer a large area of Green Belt that provides the unique setting for Oxford and contains some beautiful south Oxfordshire villages."
She added: "I want to reassure our residents we are fighting this."
Ed Turner, the deputy leader of Oxford City Council, said: "We have asked the Boundary Commission to conduct a review and it will be carried out eventually.
"We are not talking about bricks in the ground in 2009, but housing pressures are not going to go away."
