Two lanes of the southbound stretch of the motorway remain closed between junction 10, at Cherwell Valley Services, and junction nine at Bicester.
The collision happened close to junction nine shortly before 9am today.
Emergency services said they expected to reopen two remaining lanes by 1pm, but motorists have been warned to expect delays in the area.
The driver of the Land Rover had to be cut free from his vehicle and is now in the John Radcliffe Hospital in Oxford with head injuries.
His condition is described as serious, but not life-threatening.
Witnesses are urged to call police on 08458 505505 or Crimestoppers on 0800 555111.
